間接染色法如果檢查未知抗原,先用已知未標記的特異抗體(*抗體)與抗原標本進行反應,作用一定時間后,洗去未反應的抗體,再用標記的抗抗體即抗球蛋白抗體(第二抗體)與抗原標本反應,如果*步中的抗原抗體互相發(fā)生了反應,則抗體被固定或與熒光素標記的抗抗體結合,形成抗原-抗體-抗抗體復合物,再洗去未反應的標記抗抗體,在熒光顯微鏡下可見熒光。在間接染色法中,*步使用的未用熒光素標記的抗體起著雙重作用,對抗原來說起抗體的作用,對第二步的抗抗體又起抗原作用。如果檢查未知抗體則抗原標本為已知的待檢血清為*抗體,基他步驟和檢查抗原相同。
